Methodology
Transphosphorylation of p-Nitrophenyl Phosphate
Specimen Requirements
Submit only 1 of the following specimens:
PlasmaDraw blood in a green-top (lithium heparin) plasma
gel tube(s). Spin down and send 1 mL of lithium heparin plasma
refrigerated.
- Separated specimens may be stored for up to 8 hours at 25°C or for up to 7 days at 2 - 8°C.
- Freeze samples for up to 6 months at -20°C.
Note: 1. Indicate plasma on request
form.
2. Label specimen appropriately (plasma).
Serum
Draw blood in a serum gel tube(s). Spin down and send 1 mL of serum
refrigerated.
Note: 1. Indicate serum on request form.
2. Label specimen appropriately (serum).
- Separated specimens may be stored for up to 8 hours at 25°C or for up to 7 days at 2 - 8°C.
- Freeze samples for up to 6 months at -20°C.
Unspun specimens must be transported at ambient temperature and arrive to the laboratory within one hour of draw.

Interfering Substances:
EDTA when present at 200 mg/dL depresses the alkaline phosphatase results by 80 U/L at 80 U/L of alkaline phosphatase activity. Sodium citrate at 30 mg/dL increases the alkaline phosphatase activity by 25 U/L at a concentration of 55 U/L.
